Separation of Albumin from Bovine Serum Applying Ionic-Liquid-Based Aqueous Biphasic Systems

In this work, the extraction and separation of bovine serum albumin (BSA) from its original matrix, i.e., serum, was performed using a novel ionic-liquid-based aqueous biphasic system (IL-based ABS). To end, imidazolium-, phosphonium-, ammonium-based ILs, combined with anions’ acetate, arginate derived Good Buffers, were synthesized, characterized, applied in development ABS K2HPO4/KH2PO4 buffer solutions at pH 7. Initial studies commercial BSA revealed preferential migration protein to IL-rich phase, efficiencies 100% obtained single-step. recovery yields ranging between 64.0% 84.9% achieved, comprising IL tetrabutylammonium acetate leading maximum yield. With IL, directly extracted separated respective ABS. Different dilutions further investigated improve performance. Under best identified conditions, can be yield 85.6% purity 61.2%. Moreover, it is shown that secondary structure maintained process, after being phase. Overall, new herein proposed may used as an alternative platform for purification samples other added-value proteins.
